Dextran interference in biuret-based assays for serum proteins.

چکیده
We found the Easy very easy to run and without te#{233}hnique-dependent steps in the procedure. Operators need not have a high level of skill. Calibration is stable for one month, except for the ion-selective electrodes, which must be calibrated every two days. In conclusion, we found the Easy satisfactory in operational and analytical performance. The analyzer is well suited for use in the emergency laboratory, where many different persona may operate the instrument.

Elimination of dextran interference in serum protein determinations.
Serum containing dextrans of average molecular weight 40,000 (“Dextran-40”), 70,000 (“Dextran-70”), or 75,000 (“Dextran-75”) become turbid when biuret reagent is added (1, ). This interference has been overcome (3) by first precipitating serum proteins with trichioroacetic acid before applying the biuret reaction. Positive interference of these drugs with the o-toluidine glucose assay has also ...
متن کاملPrevention of interference by dextran with biuret-type assay of serum proteins.
In assay of serum proteins by use of the biuret reaction, dextran can cause turbidity by formation of an insoluble complex of dextran with copper and tartrate (or EDTA) in strongly alkaline solution. Whether or not the turbidity occurs depends on the tartrate concentration: turbidity is maximal at about 10 g/L, absent at 20 g/L or more, and only slight and delayed at 4 g/L. Two biuret reagents,...
متن کاملEffects of dextran on five biuret-based procedures for total protein in serum.
We evaluated the effect of dextran on values for total protein in serum as measured by the biuret method with five widely used automated instruments: the American Monitor Parallel; the Du Pont aca II; the Roche Cobas-Bio; the Kodak Ektachem 400; and the Beckman Astra 8. Dextran concentrations as great as 25 or 30 g/L had relatively little or no influence on total protein measurements by the lat...

A decreased A/G ratio may indicate multiple myeloma, autoimmune disease, cirrhosis, or kidney disease. The patient’s electrophoretogram was normal, and subsequent immunofixation was negative, which ruled out monoclonal gammopathy. The SPE (Fig. 1) showed an albumin of 64.4% and normal A/G ratio, which suggests spurious hyperproteinemia. The patient was being treated with dextran (1 ). In the as...
متن کاملA candidate reference method for determination of total protein in serum. I. Development and validation.
We developed a candidate Reference Method for measuring total serum protein by use of the biuret reaction. The method involves a previously described biuret reagent (Clin. Chem. 21: 1159, 1975) and Standard Reference Material (SRM) 927 bovine albumin (National Bureau of Standards) as the standard. At 25 degrees C, color development for 30 or 60 min provides identical serum protein values. Gluco...
